The ECRHS II central laboratory team based at King's College London
with the Pharmacia CAP System

Analysis of blood for specific and total IgE
The laboratory team received and maintained a serum bank from ECRHS centres where they carried out assays for serum total IgE and specific IgE for D. pteronyssinus (d1), cat (e1), timothy grass (g6) and Cladosporium (m2) using the Pharmacia CAP System and standard operating procedures. A study database of serum IgE assay results was created for central analyses.

Quality control schemes, as organised by Pharmacia and Upjohn Diagnostics and in-house, were established to ensure compatibility with methods used in the first survey

Serum samples were prepared for specific IgE tests
using the Pharmacia CAP System positioning guide


Analysis of dust
The laboratory staff· carried out house dust mite allergen assay for Der p 1 and Der f 1 and Der 2 and maintained the study database of house dust mite allergen assay results for central analyses. A quality control programme was set up consisting of solutions of known concentrations of Der p 1, Der f 1 and Der 2 included in each assay.

Mattress dust samples were sieved prior to extraction
and assay for house dust mite allergens

Microtitre plates were set up for enzyme-linked
immunoassay for house dust mite allergens


Dust samples were incubated with monoclonal antibodies
specific for 3 different house dust mite allergens
